Pathway 01
Primary (Genetic)
Caused by an inherited defect in copper metabolism. The body cannot properly excrete copper through bile, and copper accumulates in the liver over a lifetime.
- Cause
- Inherited mutation in a copper-metabolism gene. ATP7B in Labrador Retrievers and Doberman Pinschers; COMMD1 in Bedlington Terriers; suspected heritable cause in West Highland White Terriers, Dalmatians, and Skye Terriers.
- Mechanism
- Lifelong, progressive accumulation of copper in liver cells. Copper that should be packaged into bile and excreted is instead retained.
- On biopsy
- Copper accumulates in a centrilobular pattern (zone 3 of the liver lobule), typically at concentrations exceeding 1000 ppm.
- What we test
- This is the pathway the ATP7B variant is associated with, but only in breeds where the gene has been clinically linked to disease. The Chihuahua is not one of those breeds.
